3 Figure 2 Eosinophil, mast cell, and T-cell infiltration, expression of tumor necrosis factor α, epithelial cell death and apoptosis, and molecular remodeling of the epithelial layer of the esophagus in patients with EoE before and after therapy, assessed by immunofluorescence. (A) Eosinophil, mast cell, and T-cell infiltrations. Specimens were stained with anti-ECP, anti-tryptase, and anti-CD3 antibodies, respectively. (B) Expression of tumor necrosis factor α by epithelial cells. (C) Epithelial cell death and apoptosis as assessed by TUNEL assay and immunostaining using an antibody to caspase-cleaved cytokeratin 18, respectively. (D) Molecular remodeling analyses. Specimens were stained with anti-tenascin C and anti-TGF-β antibodies, respectively. All panels show representative immunofluorescence data. Appropriate control antibodies were used under each condition and showed no detectable staining (data not shown). Bars = 10 μm. Quantitative analyses for all markers are shown in Table 2. 4 Figure 3 Effect of treatment on endoscopic signs. Endoscopic pictures taken from 3 patients with histologically confirmed active EoE at baseline and after successful treatment with budesonide achieving histologic remission, showing reversibility of white exudates and red furrows but persistence of corrugated rings.
