Fig. 1 Identification of cells expressing MMP-9 by immunohistochemistry. Cell-specific staining is brown in sections incubated with sheep anti-human MMP-9 IgG and detected with HRP and diaminobenzidine shown by arrows in panel A. Nuclei are counterstained with toluidine blue. Panel B is fluorescence photomicrograph of same field as shown in panel A. Specimen was dual-labeled with monoclonal mouse anti-human CD14 IgG and detected with FITC-labeled anti-mouse IgG. Arrows indicate cells that are positive for both anti-MMP-9 IgG and anti-CD14 IgG. Black bars indicate 10 μm in magnified field. Fig. 2 Representative negative control section. AAA tissue sections were incubated with nonimmune sheep serum and processed with HRP and diaminobenzidine. Nuclei are counterstained with toluidine blue. Black bar on lower right is 10 μm in magnified field. Fig. 3 MMP-1, MMP-3, and uPA protein localization by immunohistochemistry. Sheep anti-human MMP-1 IgG labels endothelial cells (panel A) and mesenchymal cells (panel B). MMP-3 detected with sheep antihuman MMP-3 serum in mononuclear cells morphologically determined to be macrophages is shown in panels C and D. With rabbit anti-human uPA serum, uPA is detected in macrophage-like mononuclear cells in media and adventitia of aneurysmal aortic wall shown in panels E and F. Cells with positive results are indicated by arrows. Typical results with control tissues are shown at top of figure. Black bars are 10 μm in magnified fields.
