CD40L on exogenously administered anti-tumor CD8+ T cells combines with CD40L expression by endogenous CD8+ T cells to drive expression of NOS2 by monocyte-derived Tip-DCs within tumors. Production of NO− by Tip-DCs may indirectly support a T cell response or may be directly cytotoxic toward tumor cells, leading to improved survival of tumor-bearing mice treated with ACT.
