2 Figure 1 Strategy to isolate mreB and mrdA cell size mutants by flow cytometry. Cells lacking MreB or MrdA (PBP2) harbor complementing plasmids, which were exchanged for plasmids expressing mutagenized versions of mreB or mrdA (indicated by ∗). The resulting mreB or mrdA mutant strains (multicolored dots) were mixed into one library from which thinner (purple) or wider (green) cells were sorted by flow cytometry. Cells isolated after the first sort were subjected to a second sort to enrich for cell-size mutants. Normal cells (gold) were discarded.
