2 Figure 1 Programmed death ligand 1 (PD-L1) expression in 190 cases of excision specimens and paired tissue microarrays (TMAs). (A) Poor concordance of PD-L1 expression between individual excision specimen and paired TMA. Each bar chart represents one case, with the excision specimen in the red bar and paired TMA in the blue bar. Asterisk indicates cases whose PD-L1 status was assayed twice in excision specimens. (B) Scatterplot did not reveal a linear relationship between excision specimens and paired TMAs. Each dot represents one patient, and the central diagonal blue line represents identical PD-L1 expression between the two samples. (C) Distribution (%) of 190 patients according to PD-L1 tumor proportion score. 3 Figure 2 Representative images of 22C3 (programmed death ligand 1) PD-L1 immunohistochemistry (IHC) staining in excision specimens and paired tissue microarrays (TMAs). The red round area indicates the area of TMAs in the corresponding excision specimens. (A) High concordance of PD-L1 expression between TMA and excision specimen. PD-L1 expression on 100% of tumor cells in both TMA and matched excision specimen. (B) TMA overestimates PD-L1 status on the matched excision sample. PD-L1 expression on 40% of tumor cells in the excision specimen and 80% of tumor cells in the TMA. (C) TMA underestimated PD-L1 status on the matched excision sample. PD-L1 expression on 80% of tumor cells in excision specimen and negative in TMA. (D) TMA underestimated PD-L1 status on the matched excision sample. PD-L1 expression on 40% of tumor cells in excision specimen and negative in TMA. PD-L1 expression on TMAs was completely opposite to that on the corresponding area of the excision specimen.
