1 Microscopes!!!

2 How will you view cells in this class?
Using microscopes Proper way to carry microscope Carry with cord wrapped around Hold by arm and base

3 Parts of a Microscope

4 Objectives Start with scanning (lowest) objective (4X)
Move on to lower and higher powered objectives Eyepiece Ocular Lens Total Magnification Scanning (always start here!) 4X 10X 40X Low 100X High 400X

5 Learning to Use the Microscope
To view an object: Start with the stage at the LOWEST height (coarse adj knob) and the SCANNING objective in place Clip your slides in with the stage clips. Turn on the microscope. Adjust the coarse adjustment knob until you can see the sample. Adjust the fine adjustment know until the sample can be seen clearly. REMOVE the slide by turning the coarse adj knob to lowest position. When finished, wrap cord on itself! Not on microscope.

6 Making a Wet Mount Wet specimen if necessary. Place on center or well of slide. Cover with coverslip. Drop stain on one side of coverslip. Place piece of paper towel on OTHER SIDE of coverslip. Wick water through, remove slide. Place on microscope.

7 Making Scientific Drawings
Obtain a Petri dish to make “field of view”. Drawings can be in pencil/colored pencils. Must include clear, proper labels that start outside the circle. Must include magnification. Animal cells should always have- TITLE and… Cell membrane, nucleus, nuclear membrane, cytoplasm, organelles Plant cells: all above and chloroplasts, cell wall 6. Don’t just LOOK! You want to OBSERVE!

8 Quick Review Show how to properly hold a microscope.
What must you always move DOWN to begin? What OBJECTIVE must you start with to begin? What is the magnification of the ocular lens (eyepiece)? If you are viewing a specimen magnified a total of 400 X, what objective are you using?
