2 Figure 1 Immunohistochemistry of β-catenin and cyclin D1 in NSCLC. (A) Expression levels of β-catenin and cyclin D1 were analyzed using immunohistochemical staining. Representative images of positive staining are shown in adenocarcinoma (upper) and squamous cell carcinoma (lower) (×200). For β-catenin, mainly membrane and cytoplasm was stained and nuclear expression was very rare. Only cytoplasmic staining was judged as positive. Cyclin D1 was stained mainly in nucleus. (B) Prevalence of overexpression of β-catenin and cyclin D1 proteins is shown in adenocarcinoma, squamous cell carcinoma, and other cell types of NSCLC. p Values were based on Pearson’s chi-square test or Fisher’s exact test. Correlation between expression levels of β-catenin and cyclin D1 was analyzed using Spearman’s correlation coefficient in 260 adenocarcinomas (C) and in 272 squamous cell carcinomas (D). “Adenoca” and “Squamous” indicate adenocarcinoma and squamous cell carcinoma, respectively. Combined effect of β-catenin and cyclin D1 on overall survival was analyzed using the log rank test in 272 squamous cell carcinomas that were stratified by pathologic stage. Simultaneous overexpression of β-catenin and cyclin D1 was significantly associated with poor overall survival in stage IA (A) and stage IB (B), and tended to reduce the overall survival rate in stage IIA (C), but not in stages IIB (D) and IIIA (E). 4 Figure 3 The effect of β-catenin and cyclin D1 on phospho-pRb (Ser-807/811) level and Ki-67 proliferation index in stage IA–IIA squamous cell carcinomas. pRb levels phosphorylated on serine 807/811 residue (A) and Ki-67 proliferation index (B) were compared according to the expression status of β-catenin and cyclin D1. Stage IA–IIA squamous cell carcinoma patients were divided into four groups according to expression status of β-catenin and cyclin D1. Group 1 indicates patients without overexpression of β-catenin and cyclin D1. Groups 2 and 3 represent patients with overexpression of either β-catenin or cyclin D1, respectively. Group 4 consists of patients with overexpression of both β-catenin and cyclin D1. The phosphorylated pRb was stained using polyclonal anti-phospho-pRb (Ser-807/811) antibody (Cell Signaling, Danvers, MA). The phospho-pRb (Ser-807/811) levels and Ki-67 index did not show a significant difference among groups irrespective of adjuvant chemotherapy. p Values were calculated using Kruskal-Wallis test.
