What’s my total power? To calculate the total magnification, multiply the power of the ocular lens by the power of the objective. What are the powers of magnification for each of the objectives we have on our microscopes? Example: If the ocular lens has a power of10X, and the objective lens has a power of 40X, then the total magnification is 10 X 40 =400X

Comparing Powers of Magnification We can see better details with higher the powers of magnification, but we cannot see as much of the image. Which of these images would be viewed at a higher power of magnification?

The letter "e" Notice how the letter is upside down and backwards As the letter increases in size, you see less of it, but more detail 40X100X400X

1 – Place your object on a clean slide. 2 – Place ONE drop of water in the middle of the slide. Don’t use too much or the water will run off the edge and make a mess! 3 – Place the edge of the cover slip on one side of the water drop at a 45 degree angle. 4 - Slowly lower the cover slip on top of the drop. Cover Slip Lower slowly How to make a wet-mount slide …

How to focus… Begin with the stage all the way down and in low- power. Place the slide on the stage and view it first with the low-power objective. Use the coarse adjustment knob to bring the slide into focus. Once you see the image, you can rotate the nosepiece to view the slide with the high power objective. Use the fine-adjustment knob to get a clear, sharp image.

Rules… In high power, NEVER use the coarse adjustment knob! Always carry the microscope with one hand on the arm and the other under the base.
