OBJECTIVES SKILL: At the end of the practical session, the student shall be able to demonstrate the ability to estimate serum Triglycerides through quantitative method. KNOWLEDGE At the end of the session the student shall be able to explain and interpret the principle and its clinical application
Collection of sample: Serum sample is preferred. A fasting specimen is not required but may be desired to decrease Lipemia. Avoid hemolysis.
METHOD: Most commonly used method for serum triglyceride estimation is enzymatic method by glycerol-phosphate oxidase method.
Principle : Triglycerides Lipase Glycerol + Fatty Acid Glycerol + ATP Glycerokinase Glycerol 3 phosphate +ADP Glycerol-3-phosphate Glycerol Phosphate oxidase Dihydroxyacetone-phosphate +H2O2 2H2O2 + 4-Aminoantipyrine Peroxidase Quinine mine + HCl +H2O
Procedure: Take 1 cuvette. Label as Test. With the blank provided calibrate the machine. For Test mix well wait for 5 minute. Read optical density using green filter
Procedure Test Serum 20µl Reagent 2000µl
FORMULA O.D of Test x 200 = Concentration of test O.D of standard The unit is mg/dl
NORMAL VALUE Triglycerides: <200mg/dl Hypotriglyceridemia: < 60mg/dl
Cause of Hypertriglyceredimia Diabetes Mellitus Fatty Liver Alcoholism Starvation
Causes of Hypotriglyceredimia Malabsorption Syndrome PEM (protein energy malnutrition)
