Water Bath No not like the tub at home for a bubble bath! Rather a device that maintains water at a constant temperature. It is used in the laboratory for incubations 8/29/2015Property of CTE Joint Venture 5

How to Use a Water Bath Set the temperature of the water bath either with a dial control or a digital control. At the beginning of a lab or experiment, you should check the water bath to see if it is set at the right temperature and filled with water. Note: Water baths should be filled with distilled water. Centrifuge Centrifuge A piece of equipment rotates tube around a fixed axis the process causes contents to separate based on density; denser contents to the bottom of the tube lighter objects to the top. Using a Centrifuge Safety -Never place flammable liquid in a centrifuge -Always use a balance to verify weight distribution -Use correct size tube -Stay with centrifuge during start up and abort run if abnormal noise or vibration - Do not open until centrifuge has completely stopped 8/29/2015Property of CTE Joint Venture 13

Using a Centrifuge SOP Loading Samples 1. Obtain correct tubes for centrifuge you will be using and check size in centrifuge before filling tubes 2. Dispense samples into labeled tubes, cap securely, and insure tubes are in good condition 3. If needed fill water blanks into clean tubes for balancing. Use a balance and adjust the volume in your blanks to ensure even load distribution. 8/29/2015Property of CTE Joint Venture 14

Using a Centrifuge SOP Cont Inspect the centrifuge chamber prior to use to be sure it is free of any debris, dirt or leaks. Clean if needed. 5. Load samples and blanks into opposing slots or baskets. Ensure the load is fully Balanced. 6. Close chamber lid and ensure it latches properly 8/29/2015Property of CTE Joint Venture 15

Using a Centrifuge SOP Cont Set time, temp. and speed to the desired settings prior to initiating the run. 8. Remain by centrifuge until set speed is reached. If any unusual noise or vibration occurs, stop centrifuge immediately and tell teacher. 9. Close chamber lid and ensure it latches properly 8/29/2015Property of CTE Joint Venture 16
