Calibration (Qualification) of Centrifuge M. KOUPPARIS

General Successfully carry out the Preventive Maintenance Procedure and complete the Maintenance Checklist. Carry out Calibration Procedure as outlined, taking into account the specifications of the manufacturer and the methods used. This ensure that the instrument is properly maintained and calibrated to meet specifications for speed, temperature (if is controlled) and time.

Equipment Required Digital Multimeter (to measure frequency) or Optical Tachometer or Centrifuge Analyzer (to measure speed) (calibrated) Temperature Meter with temperature probe (calibrated) Stopwatch (calibrated)

Documentation Required Preventive Maintenance Procedure and Checklist (from the manual) Calibration procedure and form Calibration Label Certifications of calibration for each piece of test equipment used to perform the calibration

Preventive Maintenance Check Perform Preventive Maintenance checks as outlined in the Service Manual to ensure that the instrument is in good working order without performing any calibration Complete the Preventive Maintenance Checklist

Calibration Speed (1) Install a rotor, set the speed to 2000 rpm and start the centrifuge When the speed has stabilized, measure the actual speed with an external optical tachometer or the frequency on the multimeter, where speed = frequency x k. Record measured speed and displayed speed in the speed section of the certification form. Set the speed to the top speed of the installed rotor and after stabilization, record measured speed and displayed speed in the speed section of the certification form.

Calibration Speed (2) Repeat the check for other speed levels Calculate the %Deviation (accuracy) of rpm Evaluate the conformance to the manufacturer or method limits (e.q. 5%) and tick the pass status on the certification form. If the specification is not met, make necessary repairs or recalibration using the appropriate sections of the service manual, repeat the above speed checks and record the data in the «after repair/calibration» column of the certification form. If your are not able to repair / recalibrate call the appropriate Service Company.

Calibration Temperature (if applies) (1) Prepare samples for the rotor being used. Place the samples inside the rotor and pre-cool the samples, rotor and centrifuge chamber to 4 oC. Run the centrifuge for a minimum of 1 hr at 4 oC set temperature and a set speed equal to the top speed of the installed rotor. Stop the centrifuge and take the sample temperature using your temperature meter and probe. Record the measured temperature and displayed temperature in the temperature section of the certification form.

Calibration Temperature (if applies) (2) Determine if the temperature value recorded meet the specification of the manufacturer or the method on the form. If the specification is not met, make the necessary repairs or recalibration using the appropriate section of the Service Manual, repeat the above temperature check and record in the «after repair / calibration» column of the certification form. If your are not able to repair / recalibrate call the appropriate Service Company.

Calibration Temperature (if applies) (3) Note: A suggestion to save time would be to ask the operator to set up the samples and centrifuge to 4 oC (pre-cool) and start a hold run, the evening before, which you can shut down when you arrive and take the sample temperature. The time to pre-cool samples, rotor and chamber to 4 oC from an ambient of 25 oC, could take as long as 12 hrs.

Calibration Time (1) Using a calibrated stopwatch, check the timer by setting the time for a 10 min run. Start a manual vacuum first (if applies) When the vacuum level achieves the medium range, start the centrifuge and measure from the time the start button is pushed until the time display reads 0 and the centrifuge switches to stop.

Calibration Time (2) Record the measured time in the time section of the form. Determine if the time value recorded meets the specifications of the manufacturer or the method in the form If the specifications are not met, make the necessary repairs / recalibration or call the Service Company.

Calibration Documentation (1) After filling the certification information complete the following documentation: Log test equipment data on the calibration form Review the Preventive Maintenance checklist and calibration certification form to ensure that all data and required information has been recorded properly (including your signature and date) Fill out a calibration label and place it on the side of the centrifuge where visible

Calibration Documentation (2) Ask the operator to sign the calibration form and place a copy in Certification Record Book for that centrifuge. Place copy of the Preventive Maintenance checklist in the same Record Book Provide test equipment calibration certificates if requested by the operator / laboratory.
