ThermoSwitches Default states: ON IPTG modestly represses reporter expression Heat significantly represses reporter expression, especially for the 40C switch The OFF state is not maintained over time, however

BioSketchHarvard iGEM Speculations On the Lack of Bistability ThermoSwitch can be turned 40/37C CI is capable of repressing GFP, which is on a high-copy plasmid LacIts inactivates under heat treatment ThermoSwitch returns to basal 30C CI is not sufficiently repressing LacIts LacIts promoters are stronger than that of CI LacIts241 and 265 are very 30C than CI Collins Switch is basally 37C Can LacIts be more 30C than LacI 37C? Are there which the ThermoSwitches exhibit bistability? Are there mutations in the promoters of LacIts that affect CI repression? Mutations not present in Collins Switch plasmids nor promoter of GFP. Had to have arisen independently in both ThermoSwitches.
