ZIP13G64D protein is readily degraded by a proteasome‐dependent mechanism Proteasome inhibitor treatments. 293T cells were transfected with WT‐V5 or G64D‐V5 ZIP13 and treated with 10 μM MG132 or 1 μM bafilomycin for 6 h. ZIP13G64D protein is readily degraded by a proteasome‐dependent mechanism Proteasome inhibitor treatments. 293T cells were transfected with WT‐V5 or G64D‐V5 ZIP13 and treated with 10 μM MG132 or 1 μM bafilomycin for 6 h. Cells were lysed in 1% NP‐40 and then separated into soluble and insoluble fractions. Western blotting analysis was performed with an anti‐V5 or anti‐ubiquitin antibody. HeLa cells expressing WT‐V5 or G64D‐V5 (Supplementary Fig S2A) were treated with 10 μM MG132 for the indicated periods. (Upper) Total cell lysates were analyzed by Western blot using an anti‐V5 antibody. (Lower) The hCD8 levels indicate the amount of transfected plasmid DNA (pMX‐WT‐IRES‐hCD8 or pMX‐G64D‐IRES‐hCD8).
