Figure 6 Antagonist specifically inhibits Toll-like receptor 9 (TLR9) and Toll-like receptor 7 (TLR7) agonist-induced inflammasome activation in J774 cells. J774 cells were primed with lipopolysaccharide (LPS; 100ngml−1), TLR9 (1μgml−1), or TLR7 (50μgml−1) agonist in the presence or absence of antagonist (10μgml−1) for 4hours. Agonist-primed or resting (phosphate-buffered saline; PBS) cells were then stimulated with ATP (5mM) for 1hour. Cell lysates were processed for RNA isolation and quantitative real-time PCR analysis for NLRP3 (a) and IL-1β (b) gene expression, whereas culture supernatants were analyzed for IL-1β secretion (c).
