1 Care and use of Nikon 50i

2 Nikon 50i left side Each tick mark on the fine focus knob is measured at 1 micron

3 Nikon 50i right side Swing out condenser Fine focus wheel

4 The stage You can lower the stage and lock it in a down position. This aides in switching out specimens for study. When in down position, press down on the stage and it will unlock and raise to appropriate height

5 Adjusting brightness Viewing through eye pieces
Make sure preset switch is off (out) Rotate brightness control knob to increase or decrease brightness of image Photomicroscopy Push in preset switch Adjust brightness using either camera exposure settings or ND filter in/out

6 Diopeter adjustment Focus on specimen using 40X objective
Set 10X objective in place Hold hand over one eye and focus on image using diopeter ring Hold hand over other eye and focus on specimen using diopeter ring

7 Optical path/light distribution
Fully depressed : 100% to eyes Partially depressed: 20% to eyes – 80% to camera Fully out: 100% to camera

8 Numerical Aperture Numerical aperture number listed on objective tells you what setting to use with the aperture diaphragm.

9 4X .1 10X .25 .20 20X .40 .35 40X .65 .60 Numerical aperture
Diaphragm setting Large aperture opening increases resolution and brightness but reduces contrast and depth Small aperture opening increases contrast and depth at expense of brightness and resolution Recommended setting is .5 lower than listed numerical aperture as compromise between resolution and contrast Objective 4X .1 1 (all the way to the left 10X .25 .20 20X .40 .35 40X .65 .60

10 40X objective aperture number .65
Aperture diaphragm: .60 Aperture diaphragm: .20

11 Software

12 Annotate: This allows you to make notations and shapes on the figure and save them for future viewing Binning: If you need faster response time from camera during live viewing Exposure: If auto exposure is not giving satisfactory results, change to manual and have fun White balance: move specimen away from camera and hit Auto White while viewing the null space To capture: Hit Auto and it will capture the image and drop it into the thumbnail gallery below

13 Thumbnail gallery of captured images

14 Calibrations Camera is calibrated for all of our objectives.
Just select the objective being used with the captured image for proper calibration

15 Immersion medium We DO NOT have any objectives which can use special immersion mediums (water, glycerol or oil). Do not use these mediums with these objectives.
