4 Today’s Lab Hydrogen Peroxide
In today’s lab, we will be using chicken liver to investigate the reaction of catalase within the body. Liver cells (along with other cells in your body) produce the poisonous substance hydrogen peroxide as a by-product of cellular respiration. In order to keep the cells alive, the body uses an enzyme called catalase to break up the substance into harmless substances. This is the reaction diagram, which shows how catalase breaks up hydrogen peroxide (H202) into water (H20) and oxygen (O2).

5 Factors Affecting Reaction?
What factors could cause the rate of the reaction to change? (Increase or decrease) Think about it: person eating a hamburger What would cause him to eat (break down the meat) at a faster rate or slower rate? Now what about the enzyme catalase?

8 Lab Protocol As a group, pick one variable to test, and then design a protocol that allows you to test for that variable Plan to use 4 test tubes (one as a control) NOW: Fill out page two, write out your protocol, and check with me for the OK to get started!

9 Part B Upon completing your own experiment, carefully follow the directions for Part B on the last page, and answer the questions accordingly! When you have completely finished, make sure to clean up as described on the bottom of the last page!

18 Enzyme Limits Our results showed that as enzyme concentration increased, so did the rate of reaction Does that trend go on indefinitely? If we added TONS of enzyme, would the reaction just happen faster and faster and FASTER???

20 Enzyme Concentration We know from our experiment that as enzyme  = the reaction rate  More enzyme = more chances to collide with substrate But eventually, what happens? Reaction levels off, because not all enzymes can find substrate!

What about substrate concentration? Does the rxn rate increase go on indefinitely?? No, eventually the enzyme becomes saturated, meaning all enzymes are tied up with substrate

23 Temperature* What did the results from the experiment show, when testing cold, room temp, and heated liver? Enzymes have an ideal temperature range, because they are proteins When proteins get too hot, they denature (3D structure destroyed = unable to function), and the reaction stops
