Microscopy Observing microorganisms

Light microscopy – any microscope that uses visible light

Magnification vs. Resolution Magnification-Something that has been magnified; an enlarged representation, image, or model. Resolution- ability for lens to distinguish fine detail and structure

Total Magnification = “Magnification of specific objective lens” x “ocular lens magnification” Objective x ocular = magnification 41040 1010100 4010400 100101000

Resolution Ability of the lens to distinguish two points a specified distance apart Ex: If the microscope has a resolving power of 0.4 nm, it can distinguish two points if they are least 0.4 nm apart Light has long wavelength and cannot resolve structures smaller than about 0.2 micrometer

*To achieve high magnification with good resolution, the objective lens must be small *Specimen must contrast with substance in which they are suspended (medium) *Reflective index (light bending ability) must be changed. We do this with staining *Light rays will be refracted with the two materials and will bend * Oil immersion has same effect as increasing objective lens
