Cells were treated with the indicated concentrations of trastuzumab, lapatinib or the combination of both for 2 hours. Cell extracts were immunoblotted with the indicated antibodies. B, Activation status/phosphorylation of HER receptors in NCI-H508-HER2 and NCI-N87 cells in time-course experiments. Trastuzumab, lapatinib or the combination of both were added to cells at the beginning of the experiment at the indicated concentrations. Cells were incubated in the presence of the inhibitor(s) for the indicated times and lysates subjected to Western blot analysis. TRAS, trastuzumab; LAP, lapatinib.
