1 Expression of insulin-receptor substrate-1 and -2 in ovaries from women with insulin resistance and from controls  Xiao-Ke Wu, M.D., Ph.D., Kirsimarja Sallinen, M.D., Leena Anttila, M.D., Marjaana Mäkinen, Cheng Luo, Ph.D., Pasi Pöllänen, M.D., Ph.D., Risto Erkkola, M.D., Ph.D.  Fertility and Sterility  Volume 74, Issue 3, Pages (September 2000) DOI: /S (00)

2 Figure 1 Immunoblot detection of insulin-receptor substrate (IRS)-1 (A) and IRS-2 (B) in lysates of human cultured follicles (lane 1) and normal pregnant ovaries (lane 2). Lysates derived from a human hepatocellular carcinoma cell line (HepG2 Lysate) and mouse adipocytes served as positive controls for IRS-1 and IRS-2, respectively (lane 3). Wu. Expression of insulin-receptor substrates. Fertil Steril 2000. Fertility and Sterility  , DOI: ( /S (00) )

3 Figure 2 Immunohistochemical staining of insulin-receptor substrate (IRS)-1 (left) and IRS-2 (right) with avidin-biotin-peroxidase complex on paraffin-adjacent sections from control ovaries with normal ovulation. Each slide shows at least two different samples in each follicular stage. Increased staining for IRS-1 was found in oocytes (triangles), granulosa cells (thin arrows), and theca internal cells (thick arrows) with follicle growth from the primordial (A; magnification, ×312.5) and preantral stages (C; magnification, × 125) to the antral stage (E; magnification, ×50). Positive staining for IRS-2 in antral follicles was consistently restricted to the theca internal cells (B [magnification, ×312.5], D [magnification, ×125], and F [magnification, ×50]). Lutein cells showed intensive IRS-1 staining (G; magnification, ×125) but negative IRS-2 staining (H; magnification, ×125) . Wu. Expression of insulin-receptor substrates. Fertil Steril 2000. 4 Figure 3 Immunohistochemical staining of insulin-receptor substrate (IRS)-1 in preantral follicles (left; magnification, ×500) and IRS-2 in atretic follicles (right; magnification, ×312.5) with avidin-biotin-peroxidase complex on paraffin sections containing ovaries with four different conditions. Each slide shows at least two different samples in each follicular stage. Compared with follicles from ovulatory (A and B) ovaries or those from women with uncomplicated pregnancy (E and F), follicles in polycystic (C and D) ovaries or ovaries from women with gestational diabetes (G and H) at similar stages of development demonstrated notably increased staining for IRS-2 in the theca internal layer (thick arrows) but reduced staining for IRS-1 in the granulosa cell layer (thin arrows).
