A new approach to HIV treatment, based on the implantation of the own gene - CXCR4, CCR5 modified patient’s uninfected hematopoietic stem cells into the bone marrow by M.A. Katsin. Katsin MA, Vitebsk State Medical University (Belarus)

2 Introduction The impact of HIV infection on the human body leads to a decrease in number of functioning CD4 cells (macrophages, monocytes, T-helpers, dendritic cells). 1 per cent of human population cannot catch the HIV, because they have modified chemokine receptors (CXCR4,CCR5) on the surface of the CD4 leukocytes. I propose a new approach to the HIV treatment, based on the implantation of the own CXCR4, CCR5 modified patient’s uninfected hematopoietic stem cells into the bone marrow of the patient.

3 The 1st step of the approach
Hematopoietic stem cell isolation from the patient’s bone marrow.

4 The 2 step of the approach
Introduction of methylated primers of CXCR4/CCR5 genes into the hematopoietic stem cells. These cells will significantly limit the DNA replication inCXCR4/CCR5 genes.

5 The 3st and 4st steps of the approach
3st - Multiplication of the CXCR4/CCR5 modified hematopoietic stem cells in the cell culture. 4st – After introduction of such cells in the bone marrow, modified hematopoietic stem cells will generate all CD4 mature cells with CXCR4/CCR5 modified receptors.

6 The 6th step of the approach
Since CXCR4/CCR5 receptors are changed, HIV can’t invade CD4 modified cells, so the level of functioning CD4 cells will be sufficient, that can protect the patient’s organism from the opportunistic infection and remove different clinical manifestations of the disease with the help of effector cells (T, B cells).
