3 WHAT IS LB-MEDIA AND IT’S USES?  A substance of nutrient used for bacterial growth  An observational bed o Different types depending on utilization  Selective Media  Differential Media o LB Agar o Blood Agar PREPARATION OF LB 1. Gather all materials needed: especially Tryptone, Yeast Extract, NaCl, and Agar. 2. Calculate the amounts needed 3. Add weighed amounts to flask with half the amount of water needed. 4. Swirl and queue up to total amount. 5. Place aluminum foil over top of flask with autoclave tape. 6. Autoclave in autoclave safe bin at standard, fluid. 7. Once complete safely swirl and let cool to about 55°C. 8. Pour plates; adding ampicillin then arabinose if needed
