Microscopy

Introduction of Microscopes  Microscopic – invisible to naked eye  1595  Magnified images approximately 3x – 10x

Compound Microscopes  Uses natural or artificial light to see objects

Parts of a Compound Microscope  Ocular lens  Objective lens  Low, Medium, and High power  Body  Arm  Base  Focus  Coarse adjustment  Fine adjustment  Stage  Stage clip  Stage controls  Light source  Diaphragm

Handling Microscope  Carry the microscope with BOTH hands – one underneath the Base and one on the Arm.  Always store it with the Low Power Objective Lens above the Diaphragm.

Total Magnification  Ocular power x objective power = Total Magnification

Field of View  Measured in µm  Can be measured using ruler or graph paper

Preparing Wet Mounts  Normally use water  Starting at an angle, place coverslip gently on the specimen.  NO AIR BUBBLES

Electron Microscopes  Uses electrons instead of light  Transmission Electron Microscope  Allows user to see through object  Scanning Electron Microscope  Allows user to see the surface of object
