Effect of PD on LPS-induced activation of ERK1 and ERK2 in human monocytes. (A) Human monocytes were preincubated for 30 min with buffer or 50 μM PD , followed by a 15-min incubation with the indicated concentrations of LPS. (B) To test the effect of Ro , cells were preincubated with the indicated concentrations of Ro for 30 min at 37°C before activation with 10 ng of LPS/ml for 15 min. ERK1 and -2 activation was determined by Western blotting with an ERK1-ERK2 phosphospecific antibody. ERK1 appears as the upper band (44 kDa); ERK2 appears as the lower band (42 kDa).
