14 High frame rate photoacoustic imaging system
ultrasonic transducer Lightguide Cylindrical lens Sample This slice shows the schematic diagram of the high frame rate photoacoustic imaging system. A Q-switched Nd:YAG laser operating at 1064 nm with a pulse duration of 8 ns was used for light illumination. The laser beam was split and guided onto two multi-fiber light guides. The output beams were then focused by two cylindrical lenses resulting in two 30 mm × 3 mm light lines. Both the light guides and the focusing lenses were mounted on a fabricated holder as a part of the photoacoustic probe in order to confocally arrange the output light lines. The generated photoacoustic signals were probed by a conventional array transducer, which was also confocally mounted between the two light guides on the photoacoustic probe. The ultrasonic transducer has a pitch size of 0.3 mm and a center frequency of 5-MHz. A reflecting foil with a thickness of 9 μm was stuck on the surface of the transducer in order to shield off the backscattered laser irradiation on the transducer surface. Currently, the frame rate is limited by the pulse repetition rate of the laser system to 15 frames/s. CXCR4Ab-AuNR1000 A.R.=5.9 B Our primary gold is to prepare two types of nanoprobes: AuNR785-HER2Ab and AuNR1000-CXCR4Ab, where the subscripts represent the absorption maxima, max’s, of two kinds of gold nanorods with different particle mean aspect ratios and Ab stands for antibody. The cell lines tested in our studies include (1) the MBT-WT cells (murine bladder cancer cell line-wild type) with overexpressed HER2 and (2) human hepatoma HepG2 cells with CXCR4 overexpressed on the cell surface. The transmembrane glycoprotein human epidermal growth factor receptor 2 (HER2; p185/neu, c-ErbB-2) was also found overexpressed in breast and ovarian cancer cells. CXC-chemokine receptor 4 (CXCR4) also functions as a coreceptors for human immunodeficiency virus (HIV) and is an attractive target for the development of anti-HIV drugs. Two nanoprobes suspended in aqueous solutions were carefully prepared to exhibit well separated absorption spectral bands for our multiple targeting experiment.
