Apoptosis and proliferation of human peritoneal fibroblasts in response to hypoxia
Ghassan M Saed, Ph.D., Michael P Diamond, M.D.  Fertility and Sterility  Volume 78, Issue 1, Pages (July 2002) DOI: /S (02)03145-X

FIGURE 1 Effect of hypoxia on apoptosis of human peritoneal and adhesion fibroblast. TUNEL assay (performed as described in Materials and Methods) of untreated (A) and hypoxia-treated peritoneal fibroblasts (B) and untreated (C) and hypoxia-treated adhesion fibroblasts (D). The more green fluorescence, the greater the number of cells undergoing apoptosis. (All micrographs, original magnification, ×54.) Saed. Apoptosis of peritoneal fibroblasts. Fertil Steril 2002. Fertility and Sterility  , DOI: ( /S (02)03145-X)

FIGURE 2 Flow cytometric analysis (FACS) of apoptosis in fibroblasts from peritoneum and adhesions. FACS analysis of TUNEL assay showing that apoptosis rate is higher in fibroblasts of normal peritoneum (A, mean fluorescent channel = 18.32%) than in fibroblasts of adhesions (B, mean fluorescent channel = 14.11%) of the same patient in response to hypoxia treatment. 4 FIGURE 3 Quantification of bcl-2-bax mRNA ratio by multiplex RT-PCR. Multiplex RT-PCR of bcl-2 and bax was performed as described in Materials and Methods using RNA from peritoneum (lane 1) and adhesion (lane 2) fibroblasts. Polymerase chain reaction products were analyzed on 6% acrylamide gel and visualized by silver staining. The size of all PCR products compared with a DNA ladder (lane 3) matched the expected size for each primer pair. Saed. Apoptosis of peritoneal fibroblasts. Fertil Steril 2002. Fertility and Sterility  , DOI: ( /S (02)03145-X)

FIGURE 4 Densitometric analysis of the bcl-2-bax mRNA ratio in adhesion fibroblasts (ADF) and peritoneal fibroblasts (PF) was performed using the National Institutes of Health image analysis program. As can be seen from the graph, the bcl-2-bax mRNA ratio was approximately double in ADF as compared with PF. The higher the bcl-2-bax mRNA ratio, the lower is the apoptosis rate. Therefore, the apoptosis rate is lower in ADF than in PF. Saed. Apoptosis of peritoneal fibroblasts. Fertil Steril 2002. Fertility and Sterility  , DOI: ( /S (02)03145-X)
