2 Figure 1 NPF6.3/NRT1.1 Is a Bidirectional Transporter Involved in Nitrate Translocation from the Root to the Shoot of Arabidopsis. (A) Nitrate translocation in WT (Col-0) and chl1-5 plants. 15N content in root and shoot of WT and chl1.5 (npf6.3/nrt1.1 knockout) mutant plants is measured after different times of root incubation in 15NO3– medium. The ratio between 15N in shoot and total 15N in the plant is taken as an indicator of the nitrate translocation from root to shoot. Data are mean ± SE (n = 9–12 plants). (B) NPF6.3/NRT1.1 expression in Xenopus oocytes. Control (open circles) and NPF6.3/NRT1.1-expressing (black circles) oocytes are injected with 15NO3– to have a cytosolic nitrate concentration of 10 mM. After 0, 10, 30, and 60min, the 15N remaining in oocytes is measured. Data are expressed as percentage of 15NO3– at t = 0; n = 10–12, values are mean ± SE. (C) Specificity of nitrate efflux. 15N remaining in control, NPF6.3/NRT1.1-, NPF6.3/NRT1.1/CIPK23/CBL9-, KAT1-, NPF6.2/NRT1.4-expressing oocytes is measured at 0 (white bars) and 30min (gray bars) after 15NO3– injection. Data are expressed as percentage of 15NO3– injected at t = 0; n = 5–6, values are mean ± SE. (D) Effect of external pH on NPF6.3/NRT1.1-mediated nitrate efflux. 15N remaining in control (left) or NPF6.3/NRT1.1-expressing (right) oocytes is measured at 0 (white bars) and 30min after 15NO3– injection in external solution at pH 5.5 (dark-gray bars), pH 6.5 (gray bars), or pH 7.5 (light-gray bars). Data are expressed as percentage of 15NO3– injected at t = 0; n = 5–6, values are mean ± SE. (E) Effect of external nitrate on NPF6.3/NRT1.1-mediated nitrate efflux. 15N remaining in control (left) or NPF6.3/NRT1.1-expressing (right) oocytes is measured at 0 (white bars) and 60min after 15NO3– injection in external solution (pH 6.5) containing NO3– at 0mM (dark-gray bars) or 10mM (gray bars).
