p65 depletion increases basal protein aggregation and insolubilization. (A) Total, Triton-soluble and -insoluble protein fractions of HeLa, HeLa-Cont#1, p65-KD#1 and p65-KD#2 were separated by SDS-PAGE and analyzed in immunoblots probed with antibodies against multiubiquitin, p62 or actin (as a loading control). (B) Total protein extracts from HeLa, p65-KD#2 and p65-KD#2 cells transfected with p65FL expression vector (p65-KD#2 + p65FL). Hybridization with p65 antibody was performed for controlling the restoration of p65. (C,D) Total protein extracts of cells were prepared for filter-trap assay. Samples were then slot-blotted at four different dilutions onto a nitrocellulose membrane and probed with antibodies against multiubiquitin or p62. Mathieu Nivon et al. J Cell Sci 2012;125: © 2012.
