1 Diabetes perturbs the myeloid cell composition in wounded mice.
Diabetes perturbs the myeloid cell composition in wounded mice. CD11b+Ly6G−CCR2+ BM monocytes isolated from diabetic (Db) and non-diabetic (NDb) mouse BM flushes of animals at days 4 and 7 postwounding. (A) BM monocytes from day 7 wounded nondiabetic and diabetic mice were stained for Geimsa. Scale bar, 10 μm. (B) MFI of CCR2 and GFP on BM monocytes from nondiabetic and diabetic mice at days 4 and 7 postwounding (*p < 0.05, **p < 0.01, n = 3). Ly6C+CCR2+ cells isolated from diabetic and nondiabetic cutaneous wounds as before. (C) Representative Giemsa stains for the GFP subsets: neg/lo [(Ci) neutrophils, (Cii) monocytes, (Ciii) macrophages]; int [(Civ) monocytes, (Cv) macrophages]; and hi populations [(Cvi) monocytes, (Cvii) macrophages] from day 7 wounded diabetic mice. Scale bar, 10 μm. (D) Cell counts for the GFP neg/lo, GFP int, and GFP hi populations (*p < 0.05, n = 3). (E) Relative expression of Adgre1 and Csf1r to Hist2h2 mRNA levels, and MFI for CD11b (*p < 0.05, n = 3). Matthew Burgess et al. ImmunoHorizons 2019;3: Copyright © 2019 The Authors
