2 Fig. 1 Both the APC tumor suppressor and the cell adhesion molecule E-cadherin independently bind β-catenin. The interaction of APC with β-catenin facilitates the degradation of β-catenin, while the interaction of β-catenin with E-cadherin facilitates cell-to-cell interactions. When cytoplasmic levels of β-catenin are high, β-catenin binds architectural transcription factors of the Tcf/Lef family and in turn regulates gene expression patterns by activating and repressing target genes. β-catenin levels in the cell are tightly regulated by APC but also respond to changes in Wnt signaling. Therefore, β-catenin is a key molecule in signal transduction pathways that regulate growth, differentiation, cell-to-cell interactions, and gene expression.
