Rotation of nuclei in oocytes in the primordial follicles. (A to C) Serial images of live-imaging analysis of P7 ovaries treated with PBS (A), CTK (B), or ciliobrevin D (CD) (C). Raw and tracking images are shown (top). The lines in the right images represent trajectories. Tracking data were calculated for velocity (middle) and speed (bottom). X axes show the time points of each image acquisition (every 20 s). (D) Immunofluorescence analysis of P7 ovaries cultured for 6 hours without (control) or with CD. Images show the results of immunostaining of MVH (red) and FOXO3 (green). Nuclei were stained with DAPI (blue). Boxes in the left-hand images are enlargements from the right-hand images. Scale bars, 50 μm. The percentages of cytoplasmic FOXO3 in small (<20 μm) MVH-positive oocytes are shown. Data representing the means ± SD of three biological replicates are shown. Statistical significance was determined using Student’s t test. *P < (E) P3 ovaries at 14 days of culture without (PBS) or with CD.
