Stability of SsbA (A) and GFP (B) proteins in UA159 and ΔclpX strains. UA159 and its isogenic ΔclpX strains expressing SsbA and GFPs were grown to mid-log phase (OD600, ~0.7). Chloramphenicol was added to stop the protein synthesis, and the culture was further incubated for up to 1 h. At different time intervals, 10 ml culture was taken out and cell lysates were prepared as described in the text. Equal amounts of proteins were loaded in all lanes. The amounts of SsbA and GFP present in cell lysates were detected by Western blot analysis using anti-His antibody. BC, samples collected just before addition of chloramphenicol.
