3 Part II: Gene Expression and Cancer Poor Grandpa Joe..
Grandpa Joe (age 72), Judy Smith’s father, has been a smoker for the past 30 years. Last year, Grandpa Joe came down with a cold that turned into pneumonia. It took him more than a month to recover. Sadly, the latest tests reveal that Grandpa Joe has stage 1 adenocarcinoma, a type of non-small-cell lung cancer. The family has heard about a study being conducted at the local hospital that is looking at the genes that may be involved in the development of lung cancer. Let’s update your pedigree and Medical Intervention Chart with Grandpa Joe’s information.

4 DNA Microarray Lab… you are the research assistant!
The study is using DNA microarray technology to investigate thousands of genes thought to be involved with lung cancer. The researchers hope to compare expression of these genes of interest in lung cancer patients, to gain insight as to what causes a normal lung cell to become cancerous. Although Grandpa Joe is responding well to his cancer treatment, the family convinces him to participate in the study to help other men and women who may be at risk. You have been assigned as a research assistant to the study and will focus on five specific genes. You will use microarray technology to explore expression of the following five genes from four patients, including Grandpa Joe. Gene 1 – Human carcinoembryonic antigen (CEACAM6) Gene 2 – Surfactant protein B (SFTPB) Gene 3 – P53 tumor suppressor (TP53) Gene 4 – Sex-determining region Y (SRY) Gene 5 – Cytochrome P450 (CYP1A1)

Read the lab instructions carefully. You will need PPE (glasses and gloves). You will be working with a partner. Share the tasks…everyone should be using the micropipettor! Use the incubator—make sure you know which card is yours! After recording your results, wrap your sample card in foil to view on Friday. Write your name on the foil. Open the Google Classroom Assign. to finish.

6 What do the microarray colors mean?
What do the color mean? Make sure to compare to control samples (see table) Because the cDNA from cancerous cells is labeled with a red fluorescent marker, spots that show up as red indicate an increased expression of the gene in cancerous cells. This gene has effectively been “turned up,” or up-regulated. cDNA from healthy tissue is tagged with a green florescent dye. Therefore, spots that show up as green indicate decreased expression of the gene in cancerous cells. The gene has effectively been “turned down,” or down- regulated, in cancer cells. Spots that show up as yellow indicate equal gene expression in both cancerous and healthy tissue. Spots that show up as blank or black indicate that there was no expression of the gene in either set of cells. Sample Number Result (controls) 1 Normal Expression; equal expression in both samples 2 Increased Expression; the gene is expressed more in the cancer tissue sample than in the healthy control sample, and is said to be up-regulated. 3 Decreased expression; the gene is expressed less in the cancer tissue sample than in the healthy control sample, and is said to be down-regulated. 4 No expression; neither set of samples (cancer or healthy) expressed this gene.
