2 Figure 1 Dorsal-Skin-Resident Treg Cells Facilitate Hair Follicle Regeneration by Controlling HFSC Proliferation and Differentiation Treg cells mainly reside in primary and secondary lymphoid organs, where they function as major suppressants of inflammatory responses to promote immune tolerance. Distinct populations of Foxp3+ Treg cells can also be found in other non-lymphoid tissues, where they exert other functions. In dorsal skin, Treg cells accumulate in the HFSC niche, where they control HFSC cycling and differentiation into the keratinocyte lineage through activation of the Jag1-Notch signaling pathway. This regulation of HFSC homeostasis enables the transition from the resting telogen to the active anagen stage of the hair follicle and promotes hair regeneration at steady state and in response to depilation stress.
