3 Fig. 2 Effects of IL-1β and H2O2 on AP-1 and NF-κB DNA-binding activity. Confluent BAC were treated with H2O2 (50, 100 or 250μM) and/or IL-1β (10ng/ml). They were incubated for 3h in EBSS and treated with IL-1β or/and H2O2, then nuclear extracts were prepared (incubation 1: lanes 1–8) and a part of the cultures was further incubated for 24h in DMEM+1% FBS and treated with or without IL-1β (incubation 1+incubation 2: lines 9–17). Nuclear extracts were used to detect AP-1 (A and B) and NF-κB (C and D) DNA binding by EMSA. The binding specificity was determined using competition experiments with 50× excess of Wt or Mt AP-1 (A) or NF-κB (C) consensus probes. Inducible complexes were identified using (B): the antibodies anti-c-Fos, anti-JunB and anti-c-Jun in the presence of the consensus probe AP-1 and (D): the antibodies anti-p50, anti-p65 and anti-c-Rel in the presence of NF-κB consensus probe.
