Important advances in the fields of mouse genetics and molecular biology have helped shape our current understanding of complex systems, and we now have the tools necessary to approach challenging problems in entirely new ways. Thomas et al. show that differential enhancer usage across closely related cell types presents an opportunity for genetic manipulation. After deletion of a LyC6lo-specific enhancer region bound by PU.1, the expression of the downstream target, Nr4a1, is shut off with a concomitant decrease in levels of Ly6Clo cells. This method importantly allows for the maintenance of Nr4a1 expression in other cell types that do not depend on the developmental enhancer domain identified with whole-genome methods.
