2 Figure 1 FACS selection of Lin−CD34− cells. Blood or (BM, not shown) derived mononuclear cells were depleted of lineage-positive cells by Stem-SepTM selection. Cells were labeled with anti-CD34-APC and anti-CD2, CD3, CD15, CD19, and CD33-PE and selected on a FACS Star Plus flow cytometer. The sorting gates were set to encompass all cells within the blast window (A) and based on isotype control antibody stained samples (B). To ensure that no CD34+ cells of Lin+ cells contaminated the Lin−CD34− cell population, sort gates were drawn 1/2 log lower than the antibody control stained population (C). After FACS selection, Lin−CD34− cells were reanalyzed for presence of CD34+ or Lin+ cells. 3 Figure 2 Engraftment of 35,000 CD34+ or 35,000 Lin−CD34− from mobilized BM or blood in primary fetal sheep recipients. The 35,000 Lin−CD34− cells and CD34+ cells from blood or BM were transplanted intrauterine in 57- to 62-day-old sheep fetuses. After 2, 3, and 4 months, three to four fetuses per group were sacrificed and the percent of human CD45+ cells in the sheep BM evaluated by FACS. At 2 and 3 months, significantly more human CD45+ cells were detected in sheep that received blood-derived CD34+ cells than in the BM of recipients that received BM CD34+ cells or Lin−CD34− cells from either BM or blood. 4 Figure 3 Lower engraftment of blood than BM-derived CD34+ or Lin−CD34− in secondary and tertiary fetal sheep recipients. The 35,000 Lin−CD34− cells and CD34+ cells from blood or BM were transplanted intrauterine in 57- to 62-day-old sheep fetuses. After 2 months, three to four fetuses per group were sacrificed, and human CD45+ cells recovered from the sheep BM and equivalent numbers of human CD34+ cells transferred to secondary recipients. Secondary recipients were evaluated after 2 and 3 months. Lower human chimerism was seen in secondary recipients that received blood CD34+ of Lin−CD34− cells than BM CD34+ or Lin−CD34− cells. BM from secondary recipients sacrificed after 2 months was transferred to tertiary recipients. Blood-derived CD34+ or Lin−CD34− cells did not engraft in tertiary recipients, whereas 1% human chimerism could be detected in recipients of BM-derived CD34+ or Lin−CD34− cells Experimental Hematology  , DOI: ( /S X(00) )
