Intravital microscopy (IVM, 2-photon) imaging of ileal villi in TCRγδGFP (GFP = green channel) mice. Mice were injected with Hoechst (blue channel) before imaging to visualize all nuclei (blue). A luminal-to-crypt vertical orientation is shown. Anatomical (sub)compartments of the intestinal mucosa are shown as: Lu, lumen; IE, intraepithelial compartment; LP, lamina propria compartment. The dashed line indicates a manually drawn border. (A) Complete time-lapse showing TCRγδ+ IEL distribution in anatomical compartments over time (30 minutes). (B) Zoomed-in, higher resolution (1 μm slices) image of an ex vivo fixed ileum villus region (about 10 μm thickness), showing the location of one TCRγδ+ IEL in relation to epithelial cells in the IE compartment and the LP.
