Figure 1 Oxidized low-density lipoprotein (oxLDL), CD68, and vascular endothelial growth factor (VEGF) expression, and intimal neovascularization, in coronary artery atherosclerotic lesions of patients with estimated glomerular filtration rates (eGFRs) ≥60ml/min/1.73m2 and <30ml/min/1.73m2. (a, c, e, g, i) Serial sections of an American Heart Association (AHA) type IV lesion of a patient with an eGFR ≥60ml/min/1.73m2. (b, d, f, h, j) Serial sections of an AHA type IV lesion of a patient with an eGFR <30ml/min/1.73m2. (a, b) Light microscopic views (hematoxylin and eosin staining). (c, d) Immunohistochemical staining for oxLDL in the corresponding lesion from the boxed area indicated in panels (a) and (b), respectively. (e, f) Immunohistochemical staining for CD68 in the corresponding lesion from the boxed area indicated in panels (a) and (b), respectively. (g, h) Immunohistochemical staining for VEGF in the corresponding lesion from the boxed area indicated in panels (a) and (b), respectively. (i, j) Immunohistochemical staining for intimal CD34 in the corresponding lesion from the boxed area indicated in panels (g) and (h), respectively. Bars: in panels (a, b), 0.5mm; in panels (c–h), 0.2mm; in panels (i, j), 0.1mm. MT, Masson’s trichrome staining. Figure 2 Multivariate-adjusted mean values of positive area of oxidized low-density lipoprotein (oxLDL), monocytes/macrophages (CD68), and vascular endothelial growth factor (VEGF), and the number of intimal blood vessels, according to kidney function. (a) Multivariate-adjusted mean values of positive area of oxLDL according to kidney function were analyzed. Geometric mean values were calculated using analysis of covariance with adjustment for age, sex, hypertension, HbA1c, LDL cholesterol, high-density lipoprotein cholesterol, calcium, phosphorus, hemoglobin, smoking habits, and alcohol intake. Trends in the mean values across the estimated glomerular filtration rate (eGFR) level subgroups were tested using multiple regression analysis. (b) Multivariate-adjusted mean values of positive area of monocytes/macrophages (CD68) according to kidney function were analyzed. Geometric mean values were calculated as in the graph of panel (a). (c) Multivariate-adjusted mean values of positive area of VEGF according to kidney function were analyzed. Geometric mean values were calculated as in the graph of panel (a). (d) Multivariate-adjusted mean values of the number of intimal blood vessels (CD34-positive vessels) according to kidney function were analyzed. Geometric mean values were calculated as in the graph of panel (a). Each bar represents the geometric mean value±s.d. in each subgroup. *P<0.05, **P<0.01 vs. eGFR ≥60. Figure 3 Relationship between oxidized low-density lipoprotein (oxLDL) and vascular endothelial growth factor (VEGF) expressions and neovascularization. (a) The correlation of the expressions of oxLDL and VEGF converted to natural logarithm was analyzed (r=0.314, P<0.001). (b) The correlation of the expression of VEGF and neovascularization (mean number of CD34-positive blood vessels) converted to natural logarithm was analyzed (r=0.354, P<0.001). Figure 4 Neovascularization, intraplaque hemorrhages, and expressions of oxidized low-density lipoprotein (oxLDL) and vascular endothelial growth factor (VEGF) in an atherosclerotic lesion of a patient with an estimated glomerular filtration rate <30ml/min/1.73m2. (A) Light microscopic view of an AHA (American Heart Association) type VI lesion (Masson’s trichrome staining (MT)). Red colors in intima show intraplaque hemorrhages. (B) Corresponding lesion from the boxed area indicated in (A). (B-1) shows a high-power field of (A). Serial (B-2)–(B-4) show the immunoreactivity for oxLDL, VEGF, and CD34, respectively. (C) Corresponding lesion from the boxed area indicated in (B). Bars: in (A), 1.0mm; in (B), 0.2mm; in (C), 0.05mm.
