Column 1 (A, C, E, and G) represents the dark-field images during proliferative, secretory, menstrual phase endometrium, and sense probe control, respectively; column 2 (B, D, F, and H) represents the corresponding bright-field images. Signals for SFRP1 mRNA are detectable in all phases of endometrium and are positive in overall endometrium. However, signals are stronger in stromal area compared with glands and vessels. Note that there are signal clusters on some cells in menstrual phase endometrium. Column 3 (I and J) shows higher-magnification pictures of menstrual phase endometrium, and arrows indicate signal clusters. (b) Relative levels of SFRP1 mRNA in endometrium throughout the menstrual cycle. Quantitative PCR reactions were preformed using human endometrial tissue (n = 2) from seven different phases (n = 2). Each spot represents the relative SFRP1 mRNA level of one tissue sample. SFRP1 mRNA is detectable in all phases of human endometrium throughout the menstrual cycle. However, relative level of SFRP1 mRNA is significantly higher in proliferative-phase endometrium (Mann-Whitney test, P<.01). (c) Relative levels of SFRP1 mRNA in human endometriotic tissue (n = 6) and eutopic endometrium (n = 6). Each spot represents the relative SFRP1 mRNA level in one tissue sample. Relative level of SFRP1 mRNAs is significantly higher in ectopic tissues than in eutopic tissues (Mann-Whitney test, P<.01).
