Two broad approaches are available to recruit Tregs for therapeutic purposes. The first is to expand Tregs in vitro first and then apply expanded Tregs as a cell therapy in vivo. The advantage of this approach is that antigen-specific Tregs can be created in vitro with the use of donor-specific antigen. The second approach is to selectively and specifically stimulate Tregs in vivo by using pharmacological agents. This is accomplished by taking advantage of fundamental differences in the biology of Tregs and effector T cells. Treg, regulatory T cell.
