ENaC subunit mRNA in fresh frozen, noninflamed sigmoid colon. Biopsies of the sigmoid colon were either frozen directly after extraction or mounted into Ussing chambers and incubated in the absence of aldosterone for 8 hours. Real-time polymerase chain reaction was performed, and is expressed as fold induction of fresh frozen biopsies incubated in the absence of aldosterone. γ-ENaC mRNA levels in biopsies incubated in the absence of aldosterone were low but always detectable. In contrast, γ-ENaC could not be detected in any sample of fresh-frozen Crohn's disease colon (0/4) which is indicated as N.D. (not detectable). The results are mean values ± SEM of n = 8 controls and n = 4 Crohn's disease patients. **P < .01 compared with Crohn's disease.

AMA in Ae2a,b-deficient mice are directed against the PBC-specific pyruvate dehydrogenase complex E2 epitope. Analysis of serum immunoreactivity intensity in Ae2a,b−/− and Ae2a,b+/− mice from litters L8 and L11 at 3, 6, 12, and 15 months of age indicate that the levels of AMA (always immunoglobulin G autoantibodies) increased over time.
