Figure 5 | Ether-containing lipid biosynthesis pathways. The biosynthesis of ether-containing lipids in the peroxisome initiates the acylation of dihydroxyacetone phosphate (DHAP) with acyl-CoA to form acyl-DHAP (1), in which the acyl moiety is replaced with a fatty alcohol species to form alkyl-DHAP. Alkyl-DHAP is subsequently reduced to generate 1-alkyl glycerol-3-phosphate (G-3-P) which is acylated with acyl-CoA to produce alkylacylglycerophosphate (an ether-containing analogue of phosphatidic acid (PA)) (2). Dephosphorylation of this PA species generates alkylacylglycerol (an analogue of diacylglycerol (DAG)), which serves as a substrate for production of choline or ethanolamine glycerophospholipids (PC or PE, respectively; as shown in Fig. 4) (3). Desaturation of these ether-containing PC or PE species leads to production of plasmalogen PC or PE, respectively (4).
