Mycology revision Nada Mohamed Ahmed , MT (ASCP)i

2 Types of Culture media Type of culture media Name of the culture media
Picture NON SELECTIVE Sabouraud dextrose agar (SDA) Selective media Corn meal agar (CMA) Brain Heart Infusion (BHI) agar Bird seed agar Mycosel Agar –special for dermatophytes CHROMagar - special for Candida

3 Microscopic techniques – used to Identify of fungal morphology :
Name of techniques Method Picture wet mount(LPCB) 1-Teased Preparation gently tease the inverted side of the specimen into the staining (LPAB) fluid. 2-Scotch tape preparation touch it to the surface of the colony put it on a slide that contains a drop of lactophenol cotton blue like, 3-Slide Culture Cut the plug; put it on a sterile slide on top of the sterile water agar...

4 DERMATOPHYTOSIS (infection of dermatophytes)
Location picture Tinea corporis - small lesions occurring anywhere on the body Tinea Pedis – Athlete’s Foot Infection Tinea unguium (onychomycosis) - nails Tinea capitis - head Tinea barbae - ringworm of the bearded areas of the face and neck.

5 Diagnosis of dermatophytes (depend on Macroconidia shape)
Shape of Macroconidia picture Trichophyton Cylinder or pencil Microsporum Spindle Epidermophyton Club

6 Types of Superficial Mycoses
Causative agent diagnosis picture 1-PITYRIASIS VERSICOLOR Malassezia furfur (Lipophilic yeast) KOH- Spaghetti and meatballs 2-TINEA NIGRA Exophiala werneckii Produce melanin black or brown color Skin scrapings with KOH Septate hyphae and yeast 3-BLACK PIEDRA Piedraia hortae Septate pigmented hyphae, and asci; unicellular and fusiform ascospores. 4-WHITE PIEDRA Trichosporon. septate hyphae that develops into arthroconidia

7 Other Mycosis: Mycosis(infection) Causative agent diagnosis picture
Subcutaneous Mycosis Madurella mycetomatis Madurella grisea Demonstration of granules(graines) in the infected tissue. Systemic Mycosis Histoplasma capsulatum Giemsa or Wright stains. - Small, oval yeast like cells, 2-4µ within mononuclear or polymorphonuclear cells, narrow neck budding. Chest radiograph LPCB - White cottony mycelia with large (8-20µ) thick walled, spherical spores with tubercles or finger like projections – Tuberculate Macroconidia.

8 Candidiasis Candida. albicans KOH wet mount Fungal culture Aspergillosis Aspergillus fumigatus High level of IgE and IgG against aspergillus. Increased level of eosinophils. Sputum examination and culturePositive circulating cell wall galactomannans (are polysaccharides ) Chest X-ray.
