2 Figure 1 Two ncRNAs Promote the Assembly of Distinct Complexes on HEXIM1 (Top panel) The HDP-RNP complex is assembled on a HEXIM1/NEAT1 ncRNA platform. DNA detection detaches the HDP-RNP complex from nuclear paraspeckles. DNAPK, TBK1, and IRF3 are phosphorylated (∗), and cGAS synthesizes 2′3′ cGAMP that releases STING from the endoplasmic reticulum. STING then associates with the HDP-RNP complex, assisting IRF3 in activating the transcription of IFN genes. (Bottom panel) The HEXIM1/7SK ncRNA platform binds to and inhibits P-TEFb, thereby controlling elongation of transcription and pre-mRNA processing. 7SK titration or HEXIM1 phosphorylation increases overall P-TEFb activity.
