2 Fig. 1 Effect of partial hepatectomy on protein mass of hepatocellular organic anion transporters. Using Western blot analysis, Ntcp, Oatp1, Oatp2, Na+,K+-ATPase, Mrp2, Bsep, and ecto-ATPase were analyzed in membrane fractions isolated from liver homogenates of partial hepatectomized rats and sham-operated controls 12 hours and 1, 2, 4, 7, and 14 days after treatment. (A) Representative immunoblot. Lane 1, representative animal after sham surgery. Molecular weight (MW) markers are given in kilodaltons. (B and C) Densitometric analysis of basolateral and canalicular transporters, respectively. Autoradiographs were quantified by laser densitometry; data (mean ± SEM) are expressed as percentage of sham-operated controls. *P < No differences in protein expression were observed between animals without any surgical procedure (untreated) or sham operation at any time point examined (data not shown). 4 Fig. 2 Effect of partial hepatectomy on steady-state mRNA levels. Total RNA was isolated from partial hepatectomized rats and sham-operated controls after either 12 hours or 1, 2, 4, 7, or 14 days. Using 32P-labeled cDNAs of Ntcp, Oatp1, Oatp2, Na+,K+-ATPase, Mrp2, Bsep, and ecto-ATPase, mRNA levels were quantified by Northern blot analysis. (A) Representative autoradiograph. Lane 1, representative animal 1 day after sham surgery. (B and C) Densitometric analysis of Northern blots hybridized with cDNAs of basolateral and canalicular transporters, respectively. Autoradiographs were quantified by laser densitometry; data (mean ± SEM) are expressed as a percentage of the values obtained in sham-operated controls. *P < In case of Ntcp and Oatp2, sham operation itself reduced steady-state mRNA levels of both transporters by 40%–60% compared with untreated control animals 12 hours after surgery (data not shown). Thus, similar rates of reduction in steady-state mRNA levels are obtained when data are expressed as percentage of the appropriate sham controls. No significant changes in mRNA levels between sham-operated and untreated rats were observed at later time points for all transporter mRNA examined.
