3 Lab Safety Begins Before You Go to the Lab!
Wear gloves when handling Iodine – it is toxic. Use caution when taking your cheek cell sample – don’t stab yourself with the floss pick! Microscopes are expensive. Be nice to them. Hair needs to be tied back

4 Materials Each table group has: 1 microscope 2 glass slides
2 cover slips Forceps Flat toothpicks (for stains) Floss Pick (for cheek cell sample) Iodine Stain Bromethylene Blue Stain Elodea Sprig

5 MICROSCOPY 101 History of Microscope
Always start at lowest objective lens over the light (4x) Place microscope slide with sample centered on top of the light!! Secure with metal fasteners SLOWLY raise the stage by turning the big knob (coarse knob) Focus in once you see the cell by turning the small knob (fine focus knob)

6 Elodea Cells Place a single leaf of Elodea plant onto a glass slide
Part 1 Place a single leaf of Elodea plant onto a glass slide Add a DAB of Iodine using the flat toothpick (less is more) Place cover slip over sample Observe in the microscope under LOW power and HIGH power – draw a sketch of what you see under each magnification.

8 Cheek Cell Part 2 In each group, ONE person will swab the inside of their cheek 9-10 times using the braces floss pick. Tap your cheek cell sample onto a clean slide. Add a DAB of methylene blue stain using a flattened toothpick – less is more! Place a cover slip on the slide Observe in the microscope under LOW power and HIGH power – draw a sketch of what you see under each magnification.

10 At the End of Your Lab Time…
Dispose of slides and cover slips by placing them in the container provided by your teacher Throw gloves & used floss picks away in the trash can Wipe up any liquids (bromethylene blue or iodine) in your area- ask teacher for Clorox wipe Set up lab for next group Answer Conclusion & Questions – will be homework if you don’t finish.
