Upon oxidative stress, mitochondrial Ca2+ uniporter (MCU) senses mitochondrial ROS production through the oxidation of a matrix-facing cysteine (Cys) residue at position 97. S-glutathionylation of Cys97 induces a conformational change that promotes the clustering of MCU channels and their persistent activation. Sustained mitochondrial Ca2+ uptake decreases respiratory functions and increases mitochondrial ROS levels, leading to mitochondrial [Ca2+] overload-induced cell death.
