rSema4A potentiates Treg cells in CD4+ T cell cultures. CD4+ T cells were isolated from PBMCs by negative selection using a corresponding Ab kit and cell separation magnet obtained from STEMCELL Technologies. They were cultured in vitro with or without external rSema4A for 48 h. Then cells were harvested and assessed for the specific marker expression to detect Treg cells. The relative numbers of CD4+CD25+Foxp3+ Treg cells were selected on corresponding cell density plots in cell culture triplicates settings for each culture condition.
