Srcasm regulates levels of activated SFKs through lysosomal degradation. (1) SFKs typically are in a folded, inactive configuration. (2) Ligand binding to EGFR leads to SFK activation with phosphorylation of an active site tyrosine and binding to Srcasm through SH2- and SH3-dependent interactions. (3) Srcasm targets activated, bound SFKs for lysosomal degradation. Srcasm contains PTAP and PSAP motifs, which are putative binding sites for TSG101, a key regulator of the MVB. (4) SFKs delivered to the lysosome are downregulated by protein degradation. P, phosphotyrosine; SFK, Src-family tyrosine kinase.
