2 Figure 1 Metabolic Supply and Demand Dynamics in TSC1/2-Deficient Cells Glucose-replete cells lacking functional TSC1/2 are metabolically balanced (energy supply = energy demand) (left panel). In the absence of glucose, persistent activation of mTORC1 provokes metabolic imbalance (energy supply < energy demand), leading to energetic stress and ultimately cell death (middle panel). Manipulations that decrease ATP consumption (e.g., rapamycin treatment or inhibition of protein synthesis) restore balance (right panel) and render the cells dependent on glutamine as an alternate energy source.
